Is there a way to determine the engine size from the Serial Number on my 1955 DeSoto?

You'll have a code on your engine that identifies it. For the '55 hemi, the code is on the flat of the block right in front of the valley cover, and behind the water pump.

Original engines for '55 were 291 cid. You should find either an S21 or S22 followed by a dash and some other numbers.

It's supposed to be fun!

I'll second what Tim said, but also add that your serial number in the door jam will tell you and/or confirm if you have a Firedome or Fireflite. Firedomes were most/all 291 2 bbl engines and all Fireflites were 291 4 bbl engines from new.
