when setting ignition timing on my Desoto Firesweep 1957, shall i disconnect the vacuum advance?

Yes. Disconnect and block off vacuum source. Check point gap, warm it up, set idle rpm, set timing, reset idle rpm recheck timing. It would be really nice to get your distributor on a distributor machine to make sure your vacuum and mechanical advances are working correctly. If you have a service manual, it may refer to engine RPM or distributor RPM which is usually half engine RPM.
